Researchers at Tel Aviv University have developed a new family of drugs that may enable preventive treatment for cancer, or turn cancer into a chronic disease that can be lived with for many years.
The research, which was conducted in the laboratory of Dr. Ronit Sachi-Painero from the Department of Physiology and Pharmacology at the Sackler Faculty of Medicine, will be published in September 2010 in the scientific journal FASEB Journal.
The new research is based on the idea that the development of a cancerous tumor can be stopped by damaging the process of creating the blood vessels that supply it with oxygen and nutrients - a process known as angiogenesis. The goal of the researchers was to test the effectiveness of innovative polymeric carriers, which are designed to carry drugs directly to the cancer cell - an advanced therapeutic procedure with important advantages: the substances do not harm healthy tissues, and the amount of substance needed is significantly reduced - so that the side effects of the treatment are significantly reduced.
The carriers, developed by Dr. Ronit Sachi-Painero, Dr. Paula Ofek and their colleagues at the University of Berlin, were connected to a siRNA gene silencing agent, and the compound was injected into laboratory mice with cancerous tumors. And the results were extremely encouraging: the carriers led the siRNA directly to the cancer cells, and the compound penetrated through the cell membrane and silenced the target gene - all this without causing the mice signs of poisoning that are usually characteristic of cancer treatments. Now that the effectiveness of the procedure has been proven, the researchers hope to launch into the cancer cell siRNA responsible for silencing a key gene, which is very important in the angiogenesis process. The silencing of such a gene may inhibit the blood supply to the tumor and starve it to death. The scientists also anticipate that it will be possible to use carriers for the selective transport of additional chemotherapeutic agents, thus increasing the effectiveness of the treatment without toxicity to the patient.
According to Dr. Sacchi-Painero, this type of treatment may bring about a real revolution in the medical treatment of cancer. People who are not sick, but are at high risk of developing the disease - such as former patients or carriers of mutations in cancer-causing genes - will be able to receive preventive treatment and continue their daily routine. In cases where the cancer has already broken out, the drugs will keep the tumor in a permanent state of dormancy, and will turn the cancer into a controlled chronic disease, with which one can live for many years with a good quality of life.
Dr. Ronit Sachi-Painero won the prestigious Juludan Award for 2010 on behalf of the Technion for her contribution to the advancement of technology in the world of medicine.
