A new mechanism for artificial stimulation of the brain using ultrasound

Technion researchers have identified a new mechanism for artificial stimulation of the brain using ultrasound waves. Professors Eitan Kimmel and Shay, who are from the Faculty of Biomedical Engineering and the Russell Berry Institute for Research in Nanotechnology, tested the ability of a theoretical model that Professor Kimmel proposed three years ago - to predict the results of a series of brain stimulation experiments using ultrasound.

Creating a precise but non-invasive way to reach through the skull to target areas in the depths of the brain and influence its activity is a long-standing ambition of those involved in brain research and treatment. This is the reason for the growing enthusiasm in the scientific community for the recent successes of several research groups in the world to stimulate electrical nerve activity in nerve cells (neurons, the cells in the brain, spinal cord and nerves that communicate with each other in electrical pulses called "action potentials") using focused ultrasound beams of not high intensity. While the number of studies showing that the method works and can be applied is rapidly increasing, the stimulation mechanism remains mysterious. The new research by professors Eitan Kimmel and Shay Shao and doctoral student Michael (Misha) Flexin offers a scientific explanation for the phenomenon and may promote the ability to intervene in brain activity in a non-invasive manner using ultrasound, and lead to the development of new medical treatments. The findings were published in the scientific journal In Physical Review X

"The current work is based on a theoretical model that I proposed three years ago as a new and revolutionary explanation for the effect of ultrasound waves on cells of any type in the living body," says Professor Kimmel. "The model focused on the cellular membrane - the envelope that surrounds the cell. The molecules that make up the membrane are arranged in two layers with a space between them. According to the model, when an ultrasound wave hits the cell, the two layers of the cell membrane move apart, stretch and begin to vibrate (similar to the vibration of the vocal cords when air passes through the throat). The new research examined what happens during the vibration to the electrical behavior of the cells (the cell membrane is also a capacitor that stores an electrical charge)."

The Technion researchers showed that when the layers vibrate, the movement of the electric charge creates an alternating current that also indirectly leads to the accumulation of charge. As the vibration continues, more charge accumulates in the membrane, and eventually enough charge accumulates to create an action potential and electrical excitation of the nerve cells.

To test the model, the Technion team tested its ability to predict the results of a series of brain stimulation experiments using ultrasound. According to Professor Shaham, "a very high agreement was found between our predictions and sophisticated observations recently carried out by researchers at Stanford University. This actually has a double meaning - we developed the first theory capable of predicting neural stimulation using ultrasound, and indirectly we strengthened the theoretical model from three years ago."

This new understanding may lead to significant new medical developments. Scientists and doctors will be able to use ultrasound waves to study the internal structure of the brain non-invasively, as complementary information to an MRI scan, and possibly even to treat epileptic seizures. Professor Shoah has already begun to examine ways to stimulate cells in the retina using ultrasound - a technique that may allow the creation of images and vision without light. "There is extensive potential here for additional applications, which are based on understanding the mechanism of action," say the researchers.

    The parents are responsible for what the children see. The fact that it is convenient for parents to sit a baby for hours in front of bad programs is the source of the problem. I mean - the reason why they don't publish studies on the harms of television is the low level of television viewers - and these are the children's parents.
